The Hollywood hunk - who was recently spotted on a dinner date with newly single Jenifer Lopez - was seen getting close to Johansson when they partied together at New York's Provocateur nightspot.
"Scarlett and Bradley arrived with a small group of friends, including model Cheyenne Tozzi, around 2am, a source told LifeandStylemag.com.
"While Bradley was flirty with Cheyenne initially, after some drinks he turned his attention to Scarlett. The duo were hand in hand and more than flirty by the end of the night."
Johansson and Cooper appeared together as lovers in the 2009 film He's Just Not That Into You, and are thought to be reuniting on screen in new film Pride and Prejudice and Zombies.
Friday was the second evening running the pair spent in each other's company, after they were spotted with singer turned actor Justin Timberlake on Thursday at 1OAK bar, also in New York.
Since he split from Renee Zellweger after two years together in March, The Hangover actor Bradley has also been linked to Olivia Wilde, despite the fact he claims he has no time for romance.
He recently said: "I'm filming The Place Beyond The Pines at the moment in New York, so that's taking up a lot of my time.
"It's really exciting, but it doesn't leave much time for a relationship."
